Zieleinsatz von MIF-Familienproteinen in der Leberfibrose: Mechanismen und Translationsansätze
MIF ist ein chemokinartiges, inflammatorisches Zytokin, das mit CD74/Ii und den Chemokinrezeptoren CXCR2/4 interagiert. Unsere Vorarbeiten konnten eine mechanistisch vielschichtige, kontextabhängige Rolle von MIF bei Leberkrankheiten aufdecken, die sowohl CD74/AMPK-vermittelte hepatoprotektive Mechanismen bei toxischer Fibrose und Fettlebererkrankung, als auch pro-fibrotische Effekte in einem NASH-Modell umfasst. Im Antrag untersuchen wir die komplexen Netzwerke und neuen Mitglieder der MIF-Protein/Rezeptorfamilie (MIF-2, CXCR7, TIMP-1) und charakterisieren ihre Rolle in der Leberfibrosierung in experimentellen und translationellen Ansätzen.
